STUDY OF XANTHOMONAS CAMPESTRIS PV. CAMPESTRIS ISOLATES ORIGINATING FROM SOIL AND BRASSICA SPP. SEEDS

Xanthomonas campestris pv. (Xcc) is a harmful pathogen of the Brassicaceae agricultural plants in Serbian agroecological conditions. The bacterium survives soil, plant residues, seeds, and wild weedy plants. This study aimed to characterize Xcc isolates that persist field soil roots where they represent risk causing disease Collection samples was carried out period 2015 - 2017 four locations regions Vojvodina with cabbage cultivation monoculture or crop rotations. seed were collected same Futog Despotovo (agricultural farms) local farmer markets. All isolations done on SX YDC agar plates addition cycloheximide at 250 mg/l. Based pathogenicity tests morphological biochemical characteristics, 28 identified selected, 12 from 16 seeds. Molecular confirmation using multiplex PCR (m-PCR) Xcc-specific primer sets DLH 120/125 Zup 2309/2310. In all selected reference strain NCPPB1144, expected 370 619 bp fragments amplified. Characterizing by rep-PCR fingerprinting BOX A1R (GTG)5 primers resulted informative patterns. Cluster analysis grouped NCPPB 1144 isolates, while other tested Xanthomonads clustered separately. similarity level 90%, we obtained three representative isolates. characterization seeds will enable their comparison those pathogenic isolated leaves
